چکیده

PRIMARY non function of islet allografts and chronic failure of intrahepatic islet autografts has been observed in large animals and humans, bringing into question the liver as the ideal site for islet transplantation. Our hypothesis is that inflammatory mediators released in the microenvironment at the transplant site might result in impaired islet engraftment. The L-arginine~ependent nitric oxide (NO) pathway, which is known to inhibit rodent islet primary function.! is a putative cytotoxic mediator.

Inflammation-mediated dysfunction and apoptosis in pancreatic islet transplantation: implications for intrahepatic grafts.

Recent advances in clinical protocols have improved the outcomes of pancreatic islet transplantation (PIT), yet PIT recipients typically require pancreatic islet grafts derived from multiple donors to achieve insulin independence. This along with experimental models of syngeneic PIT, showing that up to 60% of pancreatic islet tissue undergoes apoptosis within the first several days post-transpl...

Comparison of inducible nitric oxide synthase activity in pancreatic islets of young and aged rats

Objective(s):Some pathologic situations such as diabetes and metabolic syndrome are associated with alternation in nitric oxide level. Incidence of these condition increases with aging. On the other hand, insulin secretion is modulated by nitric oxide, and nitric oxide synthase (NOS) activity is also altered in diabetes. In this study, modification in the enzyme activity associated with aging a...

Early graft failure of xenogeneic islets in NOD mice is accompanied by high levels of interleukin-1 and low levels of transforming growth factor-beta mRNA in the grafts.

Early graft failure, graft rejection, and autoimmune recurrence remain unresolved issues in islet xenotransplantation in type 1 diabetes. The first aim of this study was to examine the existence of early graft failure in spontaneously diabetic autoimmune NOD mice after rat islet transplantation under technically controlled circumstances. The second aim was to examine the mediators of this early...

CONCISE REVIEW IN MECHANISMS OF DISEASE Vascular Mediators in the Injured Liver

The vasculature plays a critical role in multiple physiologic and pathophysiologic processes. A number of recent advances have led to renewed focus on the vasculature in the liver. In patients with chronic liver disease, 3 general vascular beds are of importance. These include the peripheral vasculature, the mesenteric vascular bed, and the intrahepatic microcirculatory unit; abnormalities exis...

Complications of cirrhosis III. Hepatic encephalopathy.

Hepatic encephalopathy (HE) is a major neuropsychiatric complication of cirrhosis. HE develops slowly in cirrhotic patients, starting with altered sleep patterns and eventually progressing through asterixis to stupor and coma. Precipitating factors are common and include an oral protein load, gastrointestinal bleeding and the use of sedatives. HE is common following transjugular intrahepatic po...
